The aim of this report has been evalu ating protamine content and DNA integr ity of two patient s
with globozoospermia undergoing Intra Cytoplasmic Sperm Injection (lC SI) .
Semen analysis was carried out according to WHO criteria. Protamine deficiency and DNA
fragmentation was assessed using Chromomycin A3 and sperm chromatin dispersion assay
respectively. ICS I and chemical acti vation were carr ied out on inseminated oocyte.
Both cases demonstrated high degrees of protamine defici ency, while one of the cas es indicated
high level of DNA fragmentation, too. High fert ilization rates were achieved in both case s.
However. embr 0 transfer did not lead to implantati on or pregnancy.
Artificial oocyte act ivation overcomes low fertilization rate reported in cases with high leve l of
protamine defic iency. In the present study, fai led implantation in one of the cases may be caused
by high DNA fragmenta tion.
