Background:
The aim of this study was to establi sh a cell-free sequential culture sys tem that can
support high levels of in vitro embryo development and blastocyst format ion from bovine zygote s.
To this end, this investi gation was carried out to evalu ate the effects of glucose, serum and EDTA
on bovine zygo te in vitro development.
Materials and Methods: Bovine presumpti ve zygotes were deri ved from oocyte s matur ed, and
fert ilized in vitro and cultured in synthetic oviductal fluid sequ ential medium in a two-steps manoer;
SOF 1 for the first 3 days and SOF 2 for the second 5-6 days of in vitro embryo development. In
order to evalu ate the effect of different modi fication s of the basic medium on emb ryo developm ent,
glucos e was added to the second phase (SOF A), serum was added to the first phase (SOF C) and
EDTA alone (SOF D) or in combination with serum (SOF E) was added into the first phase of in
vitro embryo culture. The result s of each compos ition were compared with each other and with the
result s of embryo developm ent in TCM 199 vero cell co-culture system.
Results: Glucose add ition to the second phase of embryo culture, improved the devel opmental
competency; however, the differences were not significant. Serum add ition to the first phase of
embryo cultur e, significantly improved the developm ental competency of emb ryos beyond the
cleavage stage, comp ared to all the treatment andTCM 199co-culture gro ups.EDTAsupplementation
of culture medium, either alon e or in combination with serum , signifi cantly inhibits the embryo
development beyond the morul a stage.
Conclusion: The result s indicated that culture of bovine presumptive zygo tes in two steps cell-free
culture system, can support embryo development, and addition of serum through out the culture and
glu cose to the second step signifi cantly increased overall developmental competency compared to
TCM 199 co-cu lture system.
