چكيده فارسي :
بيشترين گياهان بيش انباشتگر نيكل متعلق به جنس آليسوم بوده كه بومي خاك هاي سرپنتين غرب ايران ميباشند. در اين مطالعه از گياهچه هاي A. inflatum كه به صورت هيدروپونيك كشت داده شده¬اند، براي بررسي تغييرات محتواي كلروفيل گياه تحت تاثير تركيب نيكل و كينتين استفاده شد.
چكيده لاتين :
Aim: A. inflatum is an endemic of serpentine soils from western Iran and hyperaccumulates Ni. In this
study, we used culturing callus to evaluate the effect of kinetin on SOD (EC1.15.1.1) as an anti-oxidant
enzyme engaged in resistance to metals, using a system less complicated than a developed plant both
in texture and in organ.
Material and methods: After reproducing the callus in MS medium with the concentration of 2,4-D
at 1 mg/l and 0.1 mg/l for kinetin, the callus were kept under the treatment of (0, 200, 400 μM) Ni and
(0, 0.1, 0.5, 1, 2.5 and 5 mg/l) kinetin and then the activity of antioxidant enzyme SOD was measured.
Results: The activity of SOD enzyme in the absence of Ni and at concentrations of 1, 2.5 and 5 mg/l
kinetin was increased. At 200 μM Ni concentration in the medium, none of kinetin concentrations
showed an increasing effect on enzyme activity. At 400 μM concentration of Ni in the medium, two
concentrations of 2.5 and 5 mg/l of kinetin had increasing effect on enzyme activity and as a result it
provides resistance to Ni.
Conclusion: Two concentrations of 2.5 and 5 mg/l of kinetin in the absence of Ni and also when Ni
concentration is in the range of toxicity for callus, could be considered as an activity enhancer for the
enzyme.
