Pyrazole and derivatives are key substructures in a large variety of compounds with
important biological activities and pharmacological properties [1].
Multi-component reactions (MCRs) have emerged as an attractive and powerful strategy
for organic synthesis compared to multistep reactions due to the creation of several new bonds
in a One-Pot reaction, low number of reaction and purification steps, selectivity, synthetic
convergence, high atomeconomy, simplicity, and synthetic efficiency [2].In fact, development
of MCRs can lead to new efficient syntheticmethodologies to afford many small organic
compounds in the field of modern organic, bioorganic, and medicinal chemistry [3].
We have developed a highly efficient and green One-Pot methodology for the synthesis of
a wide range of 5-amino pyrazole-4-carbonitrile derivatives, which are often encountered in
biologically and pharmacologically actives compounds. The present methodology requires
low catalyst loading as a mild Lewis acid catalyst. The most important advantages of this
method include the use of cost-effective and mild transition metal catalyst, aqueous
conditions, excellent yields, clean and simple work-up procedure, and avoidance of using
hazardous organic solvents that makes this method an instrumental alternative to the previous
methodologies for the scale-up of these One-Pot three-component reactions.
