Objectives: Closantel is a broad-spectrum antiparasitic agent used widely for the control of fasciola and haemonchus spp. infestations in sheep and cattle. The present study was carried out to compare the pharmacokinetic parameters of an oral suspension of closantel with a bolus formulation in sheep. Materials Methods: Twenty-eight male sheep, 4- 5 months of age, were randomly divided into two equal groups. Each sheep in the first group received a single dose of 10 mg/kg BW suspension, and the second group received 500 mg bolus of Closantel. The blood samples were taken from each sheep on 0, and 4, 8, 12, 16, 20, 24, 32, 48, and 72h after drug administration, and the plasma concentrations of closantel were determined using HPLC method. The Pharmacokinetic analysis was performed using a non-compartment model and the areas under the plasma concentration-time curves at 0-72h (AUC0-72), maximum plasma concentrations (Cmax) and times to reach Cmax (Tmax) of the closantel were compared in two groups.Results Conclusion: There were significant (P 0.01) differences in the AUC0-72 (2913.64± 648.14vs 1785.9±435.31µg.h/mL) and Cmax (62.133± 7.632 vs 52.367± 10.295) between suspension and bolus. How ever there was not a significant difference between two groups in terms of Tmax. The results of this study suggest that the bioavailability of oral suspension of closantel is higher than the bolus formulation.
