Studying antioxidant effects of Broccoli extract and Vitamin C on spermatogenesis and inductive oxidative stress of Diazinon

Objectives: Many of oxygen reactive species play role in infertility occurrence in males that is due to overproduction or reduction in genital antioxidant system. Organophosphates toxins are one of the causes of oxidative stress conditions. The aim of this study was to investigate the antioxidant effect of broccoli extract and Vitamin C on the spermatogenesis and inductive oxidative stress of Diazinon.Materials Methods: thirty six male Wistar rats weighing 200-250 g were randomly divided into 4 groups, including: negative control group, positive control group, first test group which received Broccoli hydro-alcoholic extract (for 300 mg/kg) in addition to Diazinon, and second test group which received Vitamin C (for 300 mg/kg). In the last day of experiment, after euthanization, Malon di-aldehyde concentration in tissue extract was measured and histological studies of testis tissue was done.Results Conclusion: Mean concentration of Malondi-aldehyde (µmol/g) was recorded as 056/0±2/2 for the first group, 126/0±42/1 for the second one, 032/0±62/2 for negative control and 017/0±8/3 for positive control. Malon di-aldehyde concentration difference between the positive control group and the first and second one was also assessed as significant. The mean sertoli, spermatogonia, spermatocyte and spermatozoid cells in the first group was recorded as 54/1±75/28, 00/8±5/91, 67/11±5/202, 59/19±184, respectively. However, in regard to the second group it was recorded as 25/1±75/25, 55/7±5/80, 33/8±31/135, 956/7±88/182, respectively. In addition to control oxidative stress conditions of Diazinon، Vitamin C - in equal dosage with Broccoli hydro-alcoholic extract - can promote spermatogenesis conditions more when comparing Vitamin C consumption alone.
