Objectives: In this study, the efficacy of PRP and the mesechyamal cells, for potentiating the tensile strength after grafting frozen allograft tendons was investigated. Design: In the present study the type of the design was experimental and analytical study. Animals: 15 four months lambs were selected for removal the both forelimbs SDF and regrafted using frozen allograft tendon of the same age. Procedures: The under strict aseptic condition mid portion of SDF of both forelimbs were removed and was grafted with allografts harvested from slaughter house and frozen under 20 degrees C0 for 60 days in all the 15 lambs, which were subsequences for grafted immediately after collections. Subsequently they were divided into 3 subgroups of 5 lambs each. The first group was acted as control ,whereas the second group 1 ml PRP was injected at the site of anastomosis and in the third group 1 ml of mesenchymal cells ( 10 6) was used at the site of grafting. After 60 days, uniaxial tensile test was done and different biomechanical parameters such as stress Fmax (N), elastic modulus (N/mm2), deformation (mm), and absorbed energy (area under load-deformation curve, N.mm) were evaluated. Results: The grafting were done under satisfactory anesthesia. In control group stress, Fmax was recorded 11.42±3.44 N, whereas in PRP group was 19.11±6.20 and in the third group it was 31.98±5.06 N. Elastic modulus was recorded 1.89±0.90 in control group whereas in second group (PRP) 3.36±0.55 and in the third group 4.20±1.90.Deformation in control group was recorded 7.00±2.41 mm and 12.66±4.90 in second group (PRP) and the third group (MSc) 16.64±4.78mm, and the last parameter recorded absorbed energy. In control group was 84.54±38.32N.mm and in PRP group 90.62±54.31 and 221.87±48.44N.mm in the third group using MSc cells. Significant changes were observed in third group (P 0.05) compared with that of PRP and control groups. Conclusion and Clinical Relevance: The results of this study showed that Mesenchymal cells PRP can besides enhancing the content of the healing tissue at the site of grafting ,it also help for early condensation of fibrous tissue at the site of anastomosis. Tensile strength evaluation is quite reliable for early normalization of grafted tendon as far as early weight bearing in the lamb.
