Introduction: Diabetes has become one of the major health issues in all societies worldwide. Exercise intervention and supplements are among the approaches to improve diabetic people’s conditions. Hence, The purpose of this study was to investigate the effects of an 8-week aerobic exercise and 25(OH)D ‌Supplementation on GLP1, DPP4 levels, fasting blood sugar, serum insulin and insulin resistance in men with type II diabetes. Methods: In this semi-experimental reserch, among 40 to 60 year old men with type II diabetes who were referred to the diabetic center of Isabn-E Maryam hospital in Isfahan of whom 48 patients were voluntarily accepted and then were randomly divided to 4 groups of : aerobic exercise group(12 patients), aerobic exercise with 25(OH)D supplement group(12 patients), 25(OH)D supplement group(12 patients) and the control group(12 patients). An aerobic exercise program was conducted for 8 weeks (3 sessions per week, each session 60 to75 minutes with 60 to 80 HR max). The supplement user group received 50000 units of oral vitamin D once weekly for 8 weeks. The GLP1, DPP4, Fasting blood sugar, serum insulin, insulin resistance and‌ 25(OH)D levels were measured before and after theintervention. At last, the data was statistically analyzed using the ANCOVA and post-hoc Test of LSD. Results: The results of ANCOVA showed a significant difference between the GLP1, DPP4, Fasting blood sugar, serum insulin and insulin resistance in aerobic exercise group and the aerobic exercise with supplement consumption group with control group, While changes in GLP1 and DPP4 levels, were not statistically significant between the supplement consumption group with control group (P 0.05). Conclusions: Aerobic exercises have resulted an increase in GLP1 and decrease in DPP4 level, fasting blood sugar, serum insulin and insulin resistance index . On the other hand, consumption of vitamin D supplement alone did not cause any changes in GLP1 and DPP4 levels, but led to an increase in 25-hydroxy vitamin D level and decrease in insulin resistance index . It seems that doing aerobic exercises with consumption of vitamin D supplement, can be used as a complementary method of treatment in improving diabetic patients’ conditions .
