60 ﻋﺪد ﻣﺎﻫﯽ ﮐﭙﻮر ﻋﻠﻔﺨﻮار ﺑﺎ وزن ﻣﺘﻮﺳﻂ 100 ﮔﺮم از ﯾﮏ ﻣﺰرﻋﻪ ﺗﺠﺎري در اﺻﻔﻬﺎن ﺗﻬﯿﻪ ﺷﺪ.ﻣﺎﻫﯽ ﻫﺎ ﺑﺎ ﻣﺤﻠﻮل 10 ﻣﯿﻠﯽ ﮔﺮم ﺑﺮ ﻟﯿﺘﺮ ﺑﻨﺰوﮐﺎﺋﯿﻦ ﻫﯿﺪروﮐﻠﺮاﯾﺪ ﺑﯿﻬﻮش ﺷﺪه و ﺑﺎ ﺻﺮف دو ﺳﺎﻋﺖ ﺣﻤﻞ و ﻧﻘﻞ ﺑﻪ ﺷﻬﺮﮐﺮد ﻣﻨﺘﻘﻞ ﺷﺪﻧﺪ اﺛﺮ ﺑﯿﻬﻮﺷﯽ ﺑﻨﺰوﮐﺎﺋﯿﻦ ﺑﺮ ﺳﻄﻮح ﮔﻠﻮﮐﺰ، ﻫﻤﺎﺗﻮﮐﺮﯾﺖ، ﮐﻮرﺗﯿﺰول ﭘﻼﺳﻤﺎ در ﮐﭙﻮر، ﭘﺲ از ﺣﻤﻞ و ﻫﻤﭽﻨﯿﻦ ﻃﯽ ﯾﮏ دوره ﺑﻬﺒﻮدي 12 ﺳﺎﻋﺘﻪ ﻣﻮرد ﺑﺮرﺳﯽ ﻗﺮار ﮔﺮﻓﺖ. ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﮐﻪ ﺳﻄﺢ ﮐﻮرﺗﯿﺰول ﭘﻼﺳﻤﺎ ﺑﺮاي ﻣﺎﻫﯿﺎن در ﮔﺮوه ﮐﻨﺘﺮل در زﻣﺎن 0 3 و 6 ﺳﺎﻋﺖ ﭘﺲ از ﺣﻤﻞ و ﻧﻘﻞ ﺑﻪ ﻃﻮر ﻣﻌﻨﯽ داري ﺑﯿﺸﺘﺮ ﺑﻮد. ﺳﻄﺢ ﮐﻮرﺗﯿﺰول ﭘﻼﺳﻤﺎ ﻣﺎﻫﯿﺎن در ﮔﺮوه ﺗﺠﺮﺑﯽ ، 3 و 6 ﺳﺎﻋﺖ ﺑﻌﺪ از ﺣﻤﻞ و ﻧﻘﻞ ﺑﻪ ﻃﻮر ﻗﺎﺑﻞ ﺗﻮﺟﻬﯽ ﺑﺎﻻﺗﺮ ﺑﻮد. 12 ﺳﺎﻋﺖ ﺑﻌﺪ از اﻧﺘﻘﺎل، ﻣﯿﺎﻧﮕﯿﻦ ﻣﻘﺎدﯾﺮ ﮐﻮرﺗﯿﺰول ﭘﻼﺳﻤﺎ ﺑﻪترتيب 62/2 ± nM 266/8 (كنترل) و 42/4 ± 162nm ( گروه تجربي) بود. به طور معني داري مقادير زيادتر ﮔﻠﻮﮐﺰ در ﮔﺮوه آزﻣﺎﯾﺶ در زﻣﺎن 0 و 1 ﺳﺎﻋﺖ ﺑﻌﺪ از ﺣﻤﻞ و ﻧﻘﻞ اﻧﺪازه ﮔﯿﺮي ﺷﺪ. 12 ﺳﺎﻋﺖ ﺑﻌﺪ از ﺣﻤﻞ و ﻧﻘﻞ، ﻣﯿﺎﻧﮕﯿﻦ ﻣﻘﺎدﯾﺮ ﮔﻠﻮﮐﺰ ﭘﻼﺳﻤﺎ 6 /. ± 12/2 mM (ﮐﻨﺘﺮل) و 0/2 ± mM 6/8 ( گروه تجربي) بود.ﻮد. ﻧﺘﯿﺠﻪ ﮔﯿﺮي ﮐﻠﯽ ﻧﺸﺎن داد ﮐﻪ، اﺳﺘﻔﺎده از ﺑﯿﻬﻮﺷﯽ ﺑﻨﺰوﮐﺎﺋﯿﻦ در ﺣﯿﻦ ﺣﻤﻞ و ﻧﻘﻞ ﻣﻨﺠﺮ ﺑﻪ ﮐﺎﻫﺶ ﻣﯿﺰان ﮐﻮرﺗﯿﺰول ﺷﺪ و ﭘﺲ از 12 ﺳﺎﻋﺖ ﺑﻬﺒﻮدي ﺳﻄﺢ ﮐﻮرﺗﯿﺰول ﭘﻼﺳﻤﺎ را ﮐﺎﻫﺶ داد ﮔﻠﻮﮐﺰ ﭘﻼﺳﻤﺎ ﭘﺲ از 12 ﺳﺎﻋﺖ ﺑﻪ ﺳﻄﺢ ﭘﺎﯾﻪ ﺑﺎز ﻧﻤﯽ ﮔﺮدد، ﮐﻪ ﻧﺸﺎن ﻣﯽ دﻫﺪ ﺣﺘﯽ ﻣﻤﮑﻦ اﺳﺖ ﺑﻬﺒﻮدي ﺑﯿﺸﺘﺮ ﺑﺮاي ﻣﺎﻫﯽ ﻫﺎ ﺑﺮاي ﺑﺎزﮔﺸﺖ ﺑﻪ وﺿﻌﯿﺖ ﻗﺒﻞ از اﺳﺘﺮس ﻻزم ﺑﺎﺷﺪ در ﻧﺘﯿﺠﻪ، در ﮐﭙﻮر ﻋﻠﻔﺨﻮار، ﺑﻨﺰوﮐﺎﺋﯿﻦ ﻫﯿﺪروﮐﻠﺮاﯾﺪ در ﻏﻠﻈﺖ 10 ﻣﯿﻠﯽ ﮔﺮم در ﻟﯿﺘﺮ ﺑﺎﻋﺚ اﻟﻘﺎء و ﺑﻬﺒﻮدي ﻣﻄﻠﻮب ﻣﯽ ﺷﻮد ﻫﻤﭽﻨﯿﻦ ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﮐﻪ ﺑﻨﺰوﮐﯿﻦ ﻫﻤﺮاه ﺑﺎ دوره ﺑﻬﺒﻮدي ﻓﺸﺎر ﻧﺎﺷﯽ از ﺣﻤﻞ و ﻧﻘﻞ را ﮐﺎﻫﺶ ﻣﯽ دﻫﺪ.
چكيده لاتين :
60 Carp of 100 g average weight were obtained from a commercial fish farm in Isfahan. The fish were anaesthetized with 10 mg/l benzocaine hydrocholoride and given a 2-h transport to Shahrekord. The effects of benzocaine aesthesia on levels of glucose, haematocrit, plasma cortisol in carps, after transport and during a 12 hours recovery period were investigated.Results showed that plasma cortisol levels were significantly higher for fish in the control group at time=0, 3, 6 hours after transport. Fish in the experimental group reached significantly higher plasma cortisol levels 3 and 6 h after transport. 12 hours after transport, mean plasma cortisol values were 266.8 ± 62.2 nM (control) and 162.0 ± 42.4 nM (experimental group). Significantly higher glucose values were measured in the experimental group at time = 0 and 1 h after transport. 12 hours after transport, mean plasma glucose values were 12.2 ± 0.6 mM (control) and 6.8 ± 0.2 mM (experimental group). In conclusion, using of benzocaine anaesthesia during transport led to a reduced release of cortisol and significantly lowers levels of plasma cortisol after 12 hours recovery period. Plasma glucose did not return to basal level after 12-hours period, indicating that even longer recovery may be needed for the fish to return to a pre-stress state. In conclusion, in grass carp, benzocaine hydrocholoride at a concentration of 10 mg/l made satisfactory induction and recovery. Also, the results showed that benzocain combined with a recovery period lessens the stress burden imposed by transport.
