Prediction of peptide binding to major histocompatibility complex (MHC) molecules is a first step for epitope vaccine development. EgA31 which is a related paramyosin protein may have value as a protective immunogen in novel vaccines against Echinococcus granolosus. This study was performed to predict potential T-cell epitopes of EgA31 protein and construction recombinant vaccine against echinococcus. Computational analysis of available EgA31 sequence was performed to find good accessibility, hydrophylicity and antigenic propensity position. T-cell epitopes were predicted using different available Server with best binding affinity which showed significantly lower 50% inhibitory concentration values. Selected peptides showed 100% population coverage. Furin linker which is enriched in Golgi apparatus where it functions to cleave proteins into their mature/active forms, consider between each peptides in synthetic gene. A partial HIV Tat sequence which is a protein transfer domain added in C-terminal of multi T-Cell epitope cassette For enhancing entery peptide to the cells. Hence synthetic DNA cloned to the Pgex vector by BamHI-XhoI double digestion and induced in BL21 cell and recombinant protein detected by Anti-GST antibody on PVDF membrane .polyepitope antigen purified by glutathione sepharose 4B. This study is first report to finding T-cell epitope in paramyosin related protein (EgA31) and construction a model of recombinant vaccine against hydatidosis.
