Enhancement of Phenolic Compounds and α-Tocopherol Production in Callus Culture of Gamay Grape Cultivar: UV-C as a Potential Elicitor
Abstract :
The present study aimed to determine the effects of UV-C irradiation on the production of some phenolic compounds (trans-resveratrol, (+)-catechin and ferulic acid) and α-tocopherol in callus cultures obtained from petiols of Gamay grape cultivar. Calli initiated from petiols cultured on the medium supplemented with 0,5 mg/L benzylaminopurine (BA) and 0.5 mg/L indole acetic acid (IAA) on B5 media, formed after 6 weeks. Callus tissues were subcultured two times each of 5 week duration. After the second subculture, 12 days old calli were exposed to UV-C irradiation. UV-C applications were carried out two different duration (5 and 10 minute) and three different distance [10cm (6,6 W/m^2), 20 cm (4,1 W/m^2) and 30 cm (2,3 W/m^2)] using UV-C lamps (30 watt) at 254 nm. Samples were collected at hours 0., 24. and 48. after UV-C treatments. The phenolic compounds and α-tocopherol were determined by the high performance liquid chromatography (HPLC). As aconclusion, phenolic compounds and α-tocopherol production in callus cultures were affected depending on UV-C treatments. While samples were exposed to UV-C at 30 cm for 10 min and collected at 48 hour showing the highest trans-resveratrol content, the combination of 5 min, 10 cm and 48 hour sampling time was the most suitable for production of (+)- catechin. The highest ferulic acid concentration was obtained from calli irradiated for 10 min at 30 cm after 24 hours. The highest α-tocopherol production was determined at samples were exposed to UV-C at 20 and 30 cm for 10min application duration and 48 hour sampling time. As a result, UV-C may be used succesfully in order to increase the production of secondary metabolites in callus cultures.
