Detection and Phylogenetic Analysis of Feline Panleukopenia Virus (FPLV) Infection in Mersin Province
Abstract :
Feline Panleukopenia (FPL) is a viral disease of cats which is usually characterized by gastroenteritis and leukopenia and it is prevalent all over the world. The virus causes infection in domestic and wild felines. In this study, 16 stool samples obtained from cats showing clinical signs of gastroenteritis were screened using conventional PCR method targeting the VP2 gene of the virus for FPLV infection and sequence analysis was performed for seven positive samples. The results of the sequence analysis were compared with the reference strains obtained from Genbank. Phylogenetic analysis showed that one sample located in G1 genotype and others located in the G3 genotype. The G1 genotype sample was closely related with FarEast strains (China, Japan and Korea), the G3 genotype samples located in the same group with Portugal and Turkey strains. The G2 genotype was not detected among the cats sampled. FPLV-positive samples were compared with reference strain (GenBank number: M38246) for amino acid changes; 1 out of 7 samples showed changes in 311.aa (D → N) and 354 aa (Y → N) and the other was 354 aa (Y → N). This study is the first report on the presence of FPLV genotypes (G1 and G3) in Mersin.
