Abstract :
As a fungal pathogen Stenecarpella maydis which leads to seedling blight and ear rot in maize is common in Africa, America, Asia and in many European countries such as Italy, France and Portugal whereas its existence has not been known in our country, yet. The only hardship for the pathogen in which the usage of healthy seed is the most significant strategy in struggling with the disease is the existence of a method that is able to provide a reliable and rapid diagnosis of the seed. The methods derived from Polymerase chain reaction, PCR, provide the determination of the pathogen before sowing, prevent inhabiting and spread of the pathogen. With this study which is carried out with this aim, the method validation concerning the determination of the isolate which is applied with the real-time PCR method and belongs to the fungal pathogen Stenocarpella maydis which takes part in EK2/A list of Plant Quarantine Regulation and is unknown in our country has been implemented. Besides, with the protocol which has been prepared, contribution to the studies of various private and official research institutions has been intended, in this way, preventing the entrance, inhabiting and spread of the pathogen has been aimed.
