EVALUATION OF ANTIBACTERIAL ACTIVITIES ON ENTEROCOCCUS FAECALIS OF CONVENTIONAL ENDODONTIC DISINFECTANTS
Abstract :
Objective: To investigate the antibacterial activity of two endodontic disinfectants (calcium hydroxide and chlorhexidine) and their combination on Enterococcus faecalis in pure in vitro conditions. Material and Method: Enterococcus faecalis A197A was grown in tryptone soy broth and the bacterial suspension was standardized. Filter paper discs with 4 mm diameter were dipped into the bacterial suspension and placed in teflon wells. Saturated solution of calcium hydroxide in water [Ca(OH)2-water] or in chlorhexidine 0.5 % [Ca(OH)2-CHX], chlorhexidine digluconate 0.5 % or phosphate-buffered saline (PBS; control) were added to wells at a volume of 75 μL. The discs were treated either for 1 or 24 hours, removed from wells, added into tubes containing 2 mL of PBS and glass beads and vortexed. Following serial dilution, 25 μL droplets were seeded onto tryptone soy agar plates. The plates were incubated at 37°C for one day and colonies were counted. Results: No bacterial growth was observed for samples exposed 1 or 24 h to disinfectants. Bacterial growth was observed in the control group; no significant difference was found between the 1 and 24 h groups (6.45±0.09 and 6.40±0.14 log10CFU/mL, respectively; p 0.05). Conclusion: In in vitro conditions, Ca(OH)2-water, CHX and Ca(OH)2-CHX combination exhibited very strong antibacterial activity
