Abstract :
Objectives: In this study, we aimed to investigate the expression levels of vascular endothelial growth factor (VEGF) which stimulates angiogenesis and vascular permeability and B-cell lymphoma-2 (Bcl-2) which plays an important role in the regulation of apoptosis in testis tissue of diabetic rats. Methods: Twenty adult Wistar albino rats were used in this study. The rats were divided into two equal groups as control and Diabetes mellitus (DM). Single dose of Streptozotosin (STZ) (55 mg/kg) dissolved in citrate buffer was injected intraperitoneally to induce DM. Tissue pieces which had been soaked in 10% formaldehyde solution were examined histologically after routine parafin follow. The expression levels of VEGF and Bcl-2 were measured by immunohistochemistry and Western Blot techniques. Results: It was observed that some spermatic cells had degeneration, nuclear contraction and picnosis in some places in DM group. While structural changes were determined in Sertoli cells, dilation and hemorrhage were observed in vessels between tubules. It was determined that VEGF expression was blocked in diabetic testis tissues, while the expression of Bcl-2 was reduced. Conclusion: Depletion of VEGF expression in diabetic testis tissues may lead to inadequate angiogenesis and vascular permeability. Moreover, a decrease in the expression of anti-apoptotic Bcl-2 in diabetic testis tissues suggests that DM may cause dysregulation of apoptosis in testicular tissue.
