Histochemical Structure of Gastrointestinal Tract of Carp (Cyprinus carpio)
Abstract :
In this study, it is aimed the determination of mucosubstance characteristic histochemical at the enlerged area anterior intestine, middle and posterior intestines of carp (Cyprinus carpio). Samples were taken from the enlerged area anterior intestine, middle and posterior intestine. All samples were fixed for 16-18 h in Bouin’s fluid. After dehydration by passing tissues through a series of alcohol solutions, the samples were vacuum embedded in paraffin. Sections (6–7 μ) were stained for general morphological purposes with haematoxylin and eosin (H and E) stains. Histochemical techniques were performed for the localization and differentiation of carbohydrate moieties In general, the histochemical staining methods applied to an examination of the reactions was found in all regions. AB pH 2.5, AF, PAS and PAS/AB pH 2.5 applicatios was stronger reaction than other applications. Mucosubstances densities increased from the enlerged area anterior intestine towards the posterior intestine. The maximum level of intensity posterior intestine was determined. AB 0.06 M ve AB 0.3 M applications quite weak reaction was observed. Mucosubstance content of carp (Cyprinus carpio) was determined to be more acidic character.
