Abstract
This study is performed to determine whether a relation exists between fusaric acid production and virulence of Fusarium oxysporum f.sp. ciceris isolates. Five isolates (KC 7, A4, KA6, KC9 ve KC6) of F. oxysporum f.sp. ciceris were grown in Czapek Dox liquid medium for 5, 10, 15 and 20 days and toxins were extracted by solid phase extraction using C18 catridge. Then, toxins were eluted with acetonitrile. The presence of fusaric acid in acetonitrile was confirmed by both thin-layer chromatography and spectrophotometer. Thin-layer chromatography of fusaric acid gave a fluoresent brick colored spot at Rf 0.70. In addition to that, an unknown metabolite was found in thin-layer chromatography analysis (Rf 0.87). Quantitave analysis of fusaric acid was performed by Agilent Specord 200 spectrophotometer. None of the isolates produced fusaric acid in the 5th day. Also, differences in fusaric acid production between low and high virulent isolates were found as statistically significant in the 20th day of incubation.
