    POLYMERASE CHAIN REACTION IN WHEAT AND OPTIMIZATION OF THE POSSIBLE PROBLEMS

  • Abstract
    Polymerase Chain Reaction (PCR) is a process of DNA amplification via DNA polymerase enzyme under artificial conditions in a thermal cycler. PCR, which has very common use in a wide range of different diciplines, has provided high speed and certainty for molecular studies. If this complex technique is used effectively after optimization, it makes studies easy for researchers. However, researcher should optimize the PCR conditions before any specific study to avoid from poor and nonspecific PCR products. The aim of this work was to put forward the problems encountered in a PCR and show how to optimize concentrations of DNA, MgCl2, dNTPs and primers. These are the critical components for a successful PCR process. By keeping other factors stable, it was determined that optimum DNA (100 ng), MgCl2, dNTPs and primer amounts were 1 μl, 2-2.25 mM, 200-400 μM and 0.25 μM, respectively. In addition, mineral oil addition to the mix had no effects on PCR product.
