Abstract
In this study, development of resistance, resistance inheritance and biochemical resistance mechanism of milbemectin in Panonychus ulmi (Koch) have been investigated. For this purpose, the susceptible population HS was also selected with milbemectin. LC_50 and LC_70 values of milbemectin in the HS population were determined primarily. As the first selection dose LC_70 value was used. Subsequent selection doses have been increased 1.5-fold each time. At the end of the fifth selection, resistance inheritance studies and biochemical studies were carried out in resistant populations. In the susceptible Panonychus ulmi population, LC_50 and LC_70 doses of milbemectin were found as 0.117 and 0.264 μl / 100 ml water, respectively. At the result of 5 selection in HS populations, the LC_50 value increased to 0.870 μl / 100 ml water and the resistance rate increased to 7.43-fold. In resistance inheritance studies, in F1 females obtained from M5 ♂ X GSS ♀ and GSS ♂X M5 ♀ reciprocal crossing that was found as D = 0.56, D = 0.06, respectively. In biochemical studies, enzymes of esterase, glutathione S-transferase (GST) and Cytochrome P450 (P450) were identified by α-naphthyl acetate, 1-chloro-2,4-dinitrobenzene (CDNB) and 7-ethoxycoumarin (7-EC) substrates. In the selection population (M5), esterase, GST and P450 enzymes increased by 1.67, 0.57 and 2.09- fold, respectively, when compared to the parental population.
