Abstract :
Platelet transfusions have a variety of adverse effects, ranging from febrile reactions to possible immuno-suppression. Some of these adverse reactions are mediated by the release of pro-inflammatory cytokines from platelets and leukocytes in platelet concentrates (PC) during blood bank storage. These cytokines appear to be released due to contact activation and micro damage of leukocytes and platelets when they come into contact with artificial surfaces of the storage container during PC preparation and storage. Storage conditions, e.g. agitation techniques, storage time and additives in PC affect cytokine release. The goal of this project is to develop modifications in PC storage conditions that will prevent the accumulation and activation of platelet-derived cytokines. To achieve this goal we are investigating the effect of modifications in PC preparation as well as different storage conditions (i.e. agitation techniques and rates) on the release of the cytokines, namely TGF-β1 and RANTES. The long-term goal is to improve the integrity and therapeutic efficacy of transfused platelets and to decrease transfusion reactions to PC.
