Abstract :
Near-infrared fluorophore pairs were comparatively studied to perform quantitative lifetime-based FRET imaging. Comparison in terms of donor lifetime reduction, relative brightness and quantitative estimation of the FRETing donor fraction were carried out using a transferrin based assay applied to breast cancer cells in vitro experiments. The best pair, as defined as the brightest and leading to accurate estimation of FRETing component is identified as the most suitable for in vitro and in vivo pre-clinical application.
