Abstract :
We have developed an all-in-one microfluidic platform that integrates sample preparation and gene analysis on one single device. Our platform takes the advantage of the silica superparamagnetic particle (SSP) based solid phase extraction (SPE) and an all-in-one scheme that performs cell lysis, DNA binding, washing, elution and polymerase chain reaction (PCR) in the same reaction chamber, which greatly reduces the complexity of the device, allowing integrating multiple reactions in parallel onto the same platform. We have successfully purify the DNA from the human whole blood sample and analyzed the Rsf gene using the Taqman probe based gene specific PCR assay on the proposed platform.
