Abstract :
Isolation and purification of anthocyanins from mulberry by column chromatography were made, macroporous resins D101 was employed to purify the anthocyanin extracts from mulberry and Sephadex LH-20 was to further separate the anthocyanins in order to obtain more purified or individual anthocyanin. HPLC-ESI/MS analysis was used to identify the anthocyanins in mulberry. The results indicated that there exist 14 anthocyanins in mulberry, mainly base on cyanidin. Three fractions were obtained from mulberry anthocyanins after purified by D101. The results showed that the major anthocyanin in fraction 1 was cyanidin-3-glucoside (85.2%) and in fraction 2 was cyanidin-3-rutinoside (78.5%), respectively. The combination of D101 macroporous resin and Sephadex LH-20 column chromatography for separation and purification of mulberry anthocyanins is an effective method, and HPLC-ESI/MS analysis is a convenient, rapid and effective method for identification of anthocyanins from mulberry.
