Abstract :
This study was aiming to investigate the fabrication and characterization of the porous chitosan/gelatin scaffold cross-linked with genipin (GP) which is widely used as a natural and non-toxic cross-linking agent. The chitosan/gelatin scaffold was prepared and cross-linked with 1% or 2%(w/w) genipin for 24 hours at 37degC, then pre-freezing for 48h at -20degC and -80degC before freeze drying. As a control, a chemical cross-linking agent glutaraldehyde (GTA) was investigated simultaneously. The swelling ratio, degradation ratio was determined by measuring change in mass between dry and wet scaffold, and measuring the change of weight in 9-days biodegradable test, respectively. Scanning electron microscopy (SEM) was used to observe the scaffold morphology. The XTT assay was applied to evaluate the cytotoxicity and biocompatibility of the chitosan/gelatin scaffold with MC3T3-E1 cells in this study.
