  • Abstract
    The trehalase (TRE) is one of enzymes related to trehalose catabolism in plant. On the basis of previous experiments, we are trying to inhibit the expression of trehalase gene through RNA interference technology in order to achieve the purpose of increasing the accumulation of trehalose in plant. So the fragments of TRE gene and RNAi-TRE have been obtained from Arabidopsis seedling by using RT-PCR (reverse transcription PCR) method, then they were constructed into pBI121 (plant expression vector) and pGSA1285 (RNAi expression vector) respectively. After transformed to tobacco, the expression of TRE gene has been detected through real-time quantitative PCR in transgenic tobacco. The result shows that the mRNA level of TRE gene decreased obviously in transgenic tobacco which has been transformed with RNAi-TRE. It indicated that RNA interference technology is also effective to heterogeneous plant in regulation of gene expression.
