Abstract :
Interferen-γ of Rongchang porcine was amplifed by RT-PCR with the total RNA extracted from the peripheral blood lymphocytes. The gene was sequenced after cloned into the vector pMD18-T and analyzed with molecular biological software. The results showed that the opening reading frame (ORF) of interferen-γ of Rongchang porcine consists of 501 nucleotides, encoding 166 amino acids, the front 23 amino acide is signal peptide. The homology of IFN-γ of rongchang porcine with chenghua porcine interferon is 99.8% at nucleotide level. The homology of susscrofa IFN-γ is 96.8%. the homology of rongchang porcine IFN-γ with that of camel, rabbit, horse, goat, cattle, gorilla and human is 84.8%, 71.8%, 77.6%, 60.7%, 84.0%, 69.5% and 70.3%. Phylogenetic analysis based on IFN-γ gene showed that rongchang porcine had the closest linkage to chenghua procine.
