Abstract :
A novel Or83b was cloned from antennae of Heliothis viriplaca (Lepidoptera: Noctuoidea) by using RT-PCR and RACE methods, which was named HvirOr83b (GenBank accession no. JQ394904). The cloning and sequencing results showed that the full-length of open reading frame in HvirOr83b was 1422 bp, encoding 473 amino acid residues with seven transmembrane domains, which shared typical feature of Or83b with those of other insects. The HvirOr83b protein has a deduced molecular weight of 53.5 kDa and an isoelectric point of 8.37. Blast searching in NCBI showed that the deduced amino acid sequence was highly similar to that of other Or83bs from Lepidoptera with acid sequence identities of 62%~99%, and the HvirOr83b was remarkably conserved among many insects and can be used to unveil speciation and the direction of differentiation in closely related species. It is showed that the HvirOr83b was observed in the antennae, abdomen and legs of male and female by tissues expression analysis.
