Abstract :
An ultrasonic-wave assisted technique for the extraction of water-soluble polysaccharides from Atractylodes Macrocephala Koidz have been developed based on orthogonal test method. A comparison of polysccharides extraction yield between ultrasonic wave extraction and the conventional boiled water reflux method has been investigated. The orthogonal test was employed to test the effects of volume of water, time of extraction, temperature of extraction, and times of operation on the yield of polysccharides. Four convenient methods for protein removal were employed purification of polysaccharides. The new processing was feasible to operate, and had the advantage of yield of polysaccharides than convention methods. The optimal process parameters were as follows. 12 times volume of water was performed, time of extraction was 60 min, temperature of extraction was 60degC, and times of operation was twice. The best method of protein removal by lead acetate was adopted.
