Sequence Characterization and Molecular Detection of the 3´-Terminal Region of Bean Yellow Mosaic Virus Infecting Canna

Abstract :
The first sequence obtained from a Potyvirus infecting Canna plants (Canna spp.) revealed that it is a new strain of Bean yellow mosaic virus (BYMV), without noticeable recombination with other plant RNA viruses from this region being found. Using a degenerate primer for all Potyvirus, a DNA fragment of about 1.6kb size was amplified from total RNA samples extracted from natural-diseased Canna plants. Results of multiple alignments showed that CP nucleotide sequence, its deduced amino acid sequence and 3´ UTR sequence of BYMV Canna isolate (BYMVHZCanna) had homologies of 81.5%~85.8%, 84.7%~91.6% and 93.0%~95.9% to those of other isolates recorded by GenBank. BYMVHZCanna alone was classified in a cluster in three phylogenetic trees, which were based on CP nucleotide sequences, its deduced amino acid sequences and 3´ UTR sequences of all BYMV isolates. Field survey with RT-PCR detection showed that this virus has been widely spread in all the surveyed areas such as Hangzhou, Guangzhou and Shenzhen cities, in China.
