Abstract
Study on microbial community structure and diversity in freshwater is important for exploiting the relationship of microbial community and its habitats, predicting and monitoring the ecological changing of contaminated water habitat. The retrieval of 16SrDNA genes directly from the environment has made it possible to estimate the phylogenetic diversity of a natural bacterial community without preceding cultivation steps, which reflect more comprehensively of bacterial community in the water samples in different ecological restoration areas. The genomic DNA was extracted from water, PCR amplification of this gene fragments; DGGE was analyzed; the abundance of bacteria were observed. The result showed that the bacterial community and abundance changed in different ecological restoration sections. With the depth of bio-disposal, the diversity of microbe community increased, however, the abundance of bacteria declined, which demonstrated the ecological restoration engineering improved the water habitat, promoted the microbe community diversity in the eco-restoration areas.
