Abstract
In order to enhance cell and spore concertration of Bacillus Licheniformis SH003, the optimum fermentation conditions of Bacillus Licheniformis SH003 were studied by single-factor test in shake flasks conditions. The results indicated that culture conditions were: agar volume 40mL, inoculum concentration 10%, initial pH 6.5, growth temperature 35°C, fermentation time 36h and shaker rotating speed 220r/min. According to the nutritional needs of Bacillus Licheniformis SH003, basis culture medium was optimized by single-factor test and orthogonal test. The result showed that the optimizational culture medium had the composition (W/V): sucrose 3.0%, peptone 1.0%, K2HPO4 0.2%, MnSO4 0.005% and MgSO4·7H2O 0.1%. In the end, the effects of 20 kinds of traditional Chinese herbs, such as Pericarpium citri reticulatae, as promoting factors on the growth of Bacillus Licheniformis SH003 were investigated. The results showed that 0.9% (W/V) Kaempferia galanga L. extract had the greatest influence on the growth of Bacillus Licheniformis SH003. Growth curve of Bacillus Licheniformis SH003, which were fermented with optimum multiplication culture condition, was measured. In stationary phase, CFU was 3.1 × 109cfu/ mL, and sporulation rate was 64.3%, which were as much as 13.5 times and 2.41 times of having no optimized multiplication culture medium.
