Abstract :
In order to analysis of chlorogenic acid in Do-Zhong(Eucommia ulmoides), ultrasolic extraction was used to prepare the sample. With yield of chlorogenic acid as index. The effect of ethanol concentration, ultrasolic power, extraction time, and the ratio of solid to liquid were studied. The extracts were then analysis by high-performance liquid chromatographic (HPLC) method. The results shown that the optimum extraction conditions were as follows: 70% ethanol(w/w), extraction power 200W, extraction time 18 min, stock ratio 1:10(g:mL). For HPLC analysis, the calibration plot was linear (R2=0.9993) in the concentration range 200~1000μg/mL for chlorogenic acid. The method was precise with relative standard deviation for chlorogenic acid range between 1.50~2.55%. The recovery of this method were between 98.46 -100.05%. The method could be used as a practical technique for detect chlorogenic acid in Do-Zhong for it was samples rapid, simple, sensitive, convenient..
