Abstract :
Temporally and spatially regulating the microtubule cytoskeleton, the actin cytoskeleton and cortical myosin assembly is primary to cell dividing. To realize the contribution of actin and microtubule in polar cortex at early anaphase, we locally released cytochalasin D and colchicine in a highly localized manner to polar region of dividing normal rat kidney cells. The results showed that local release of CD or colchicine at the polar region caused inhibition of cytokinesis at early anphase before ingression. Cells treated with 30μM blebbistatin were used to contrast to control. Our findings showed that disturbed polar cortical cytoskeleton not only inhibited initiation of cytokinesis but also influenced the position of furrow. The results suggested the definitive roles of global coordination of cortical activities at early anaphase.
