Abstract :
CsCCD8 was a homologous gene of inhibiting axillary bud growth in cucumber and cloned by in silico cloning technique. Utilizing a series bioinformatics softwares, database and online programs, CsCCD8 nucleotide sequence and its amino acid sequence were analyzed and found that CsCCD8 cDNA full sequence consisted of 1 641 nucleotides and encoded a 546 AA which was homologous with carotenoid cleavage dioxygenase 8 in Arabidopsis thaliana. After blasting in cucumber genomic database, CsCCD8 was much more homologous with Csa018589 at Chromosome 2 in cucumber, full length genomic sequence was 7 035 bp and included totally six extrons and five introns in this genomic sequence. CsCCD8 molecular weight was 60.6KDa and was an stable protein. There are so many phosphorylation sites in CsCCD8. CsCCD8 was located in chloroplast. The CsCCD8 secondary structure contained several kinds of conformations, and was mixed type. The results of predicting secondary and tertiary structures showed that CsCCD8 was classified into carotenoid oxygenase family. CsCCD8 was belonged to CCD8 evolution branch by phylogenetic analysis.
