Phellinus mushrooms can be easily identified to genus level by morphological characters; however, the taxonomy of species is very confused. The sclerotium producing mushroom, Hypholoma tuberosum is not recorded occurring in China. In this paper, the internal transcribed sequence (ITS) of two wild occurring mushrooms, P. igniarius CBL and H tuberosum TUB2, were assayed, and the phylogenetic tree of both genus based on sequences of ITS1+5.8S+ITS2 were built. The results suggested that at level of 95% identity, eighteen materials of genus Phellinus were clustered into two groups and the tested strain of P. igniarius CBL was in group II. Moreover, it can be concluded from the phylogenetic tree that genus Phellinus is probably polyphyletic. All species of genus Hypholoma were clustered together at level of 96% similarity, and the molecular taxonomy of the tested strain of H. tuberosum TUB2 was consistent with that based on morphological characters. The assay of ITS sequence of fungal rDNA is simple and fast, and thus the sequence analysis will be very helpful to many mycological researchers who are not professional taxonomist in identification and further exploitation of wild occurring mushrooms.
