The orthogonal matrix method with three factors each at three levels was employed to optimize the extraction of polysacchride from Bombyx batryticatus. Optimization results show that the optimum ultrasonic treatment time was 50 min, extraction temperature 100°C and extraction time 1 h. The effects of different granularity on extraction were comparatively studied. The results showed that with the optimized condition the Bombyx batryticatus milled through 160 size mesh produced optimal results, whose polysaccharides content reached high up to 11.55±0.69 mg/g. Content changes of polysaccharides were also investigated in the silkworm bodies after surface inoculation by Beauveria bassiana. The results showed that polysaccharides content had increased significantly from 2.07±0.24 up to 11.84±0.44 mg/g during 7 days.
