It is well known that tumor vaccines induce protective humoral and cell-mediated immune responses in several animal models .In this study, we took the S180 ascites lump cells treated with polysaccharide isolated from porcine cartilage for 48h as a tumor vaccine. The goal of this study was to investigate whether this tumor vaccine is able to augment the antitumor immune responses in a mouse modeLand investigate the immunologic mechanism of the antitumor immune responses. We previously demonstrated that cartilage polysaccharide significantly inhibited tumor growth and induced apoptosis by MTT assay, analysis of cell cycle and hoechst 33342/pi staining in vitro, and sequentially demonstrated that the mice immunized with the tumor vaccine were suppressed tumor growth and extended the survival rate by immune experiment in vivo, followed by we demonstrated that the tumor vaccine enhanced the antitumor immune responses by Immunofluorescence staining, finally, we identified the reactive proteins extracted from untreated s180 cells and the pretreated S180 cells by Western blot and 2D-PAGE, we found the expression of a protein at an estimated molecular mass of 35kDa was increased after treatment with Cartilage Polysaccharide. These results indicate that the structure and expression of tumor antigen in S180 cells were changed in response to a treatment with Cartilage Polysaccharide. Therefore, the anti-tumor immunity of mice were activated by the tumor vaccine so that the mice immunized by the tumor vaccine could prevent fr- m invasion of tumor cells.
