Abstract :
Effects of spacer arm heterology on antibody production, screening and ELISA was investigated with the help of computational molecular modeling. Three chloramphenicol (CAP) haptens with different spacer arms were conjugated with carrier proteins. Balb/c mice was immunized with complete antigen, then serum titre and affinity were detected. Theoretic geometric and electronic parameters were estimated, the similiarity between haptens and CAP were compared. A new CAP immunogen with para-methyl benzoate as spacer arm was synthesised. Spacer arm with para-oriented aromatic ring induced extreme structural change of haptens, yet improved the sensitivity of ELISA test.
