In our previous study, we have found that the intracellular reactive oxygen species (iROS) to mediate the monocytic differentiation in leukemia cells (HL-60) induced by isoliquiritigennin(ISL), NADPH oxidase is the main source of reactive oxygen species in the cell. In order to find out the relationship between ISL-induced differentiation and NADPH oxidases, the content of Superoxide anions (O2-) was detected. ISL increased intracellular Superoxide anions (O2-) that was completely inhibited by the NADPH oxidase inhibitor DPI and APO. ISL increased NBT reduction ability and the expressions of surface antigens CDllb were completely inhibited by DPI and APO. Our data suggest that NADPH oxidase-mediated generation of ROS is an important contributor to ISL-induced HL-60 cell differentiation.
