Comparative study on callus induction from different explants of T.lutarioriparia L.Liu and chemical imaging of vesicular structure by confocal microscopy

Abstract :
To establish the optimal regeneration system of T.lutarioriparia L. Liu, several induction-media with different plant growth regulators concentrations were orthogonally designed to induce callus from different explants of T.lutarioriparia. L.Liu. The results showed that 2, 4-D is important for the induction of callus from explants of roots, immature inflorescences and mature seeds. However, KT and 6-BA are the key factors to the induction of callus from explants of young shoots. Calli are found to emerge after 7-10d, 15-20d, and 20-25d, respectively. The calli induced from various explants have different callus induction frequency (CIF) and callus differentiation frequency (CDF). The CIF of immature inflorescences, young shoots, seeds and young roots were 96%, 42%, 37.5% and 5.72%, respectively. The CDF of immature inflorescences, young shoots and seeds were 100%, 85% and 80%, respectively. The highest number of seedlings per callus were regenerated after the third subculture. Vascular bundles were observed by confocal microscopy from young stem of plants, but only tracheids were observed from callus.
