Abstract
In order to improve the extraction technology of Polysaccharides from Ophiocordyceps sobolifera and its antioxidant activity, and an optimal enzyme method combined response surface methodology was provided in this paper. Take the Polysaccharides extraction rate processed by protease, cellulase and pectase as response value, while the amount of enzyme, enzymolysis temperature, enzymolysis time, and its pH as response factors, the experimental design of four factors and three levels was applied to Box-Behnken central composite method. The antioxidant activity of the extracts was determined by spectrophotometry. The optimal conditions for the extraction were found to be: the amount of enzyme of 1.5%, enzymolysis temperature at 49?, enzymolysis time of 150 min, pH at 4.8 and ratio of solid to solvent of 1:20 for Ophiocordyceps sobolifera Polysaccharides. Regression analysis showed that 1.2% relative error between experimental values and the model employed. The elimination of hydroxide free radical was at 44.51%. These indicated that optimization of extracting technology condition be reliable, which based on response surface method, and the extraction has a high antioxidant activity.
