بررسي مقايسه اثرات ضدقارچي عصاره‌‌هاي مختلف گياه onosma chlorotricum بر قارچ‌هاي كانديدا آلبيكنس و كانديدا گلابراتا با دو آنتي‌بيوتيك فلوكونازول و نيستاتين

Comparison of the antifungal effects of various extracts of Onosma Chlorotricum on Candida albicans and Candida glaberata with two antibiotics fluconazole and nystatin

با توجه به عود عفونت هاي كانديدائي و مقاومت آن ها به داروهاي ضدقارچي در طي درمان و نيز عوارض شناخته شده داروهاي شيميايي و وجود بعضي تحقيقات مبني بر اينكه جنس onosma حاوي مشتقات آلكانين و شيكونين بوده كه داراي خواص ضدقارچي مي باشند، براي اولين بار خواص ضد قارچي گياه onosma chlorotricum در مقايسه با آنتي بيوتيك مورد بررسي قرار گرفت.مواد و روش ها: گياه مذكور در اواسط تيرماه از شهرستان كوهدشت جمع آوري و پس از شناسايي و خشك كردن، عصاره هاي انهگزاني، متانولي و آبي به روش خيساندن تهيه شد. اثرات ضدقارچي غلظت هاي مختلف عصاره ها با استفاده از روش انتشار ديسك و mic (حداقل غلظت ممانعت از رشد)، mfc (حداقل غلظت قارچ كشي) به روش ميكرودايلوشن بررسي شد. آنتي بيوتيك فلوكونازول و نيستاتين به عنوان كنترل مثبت، dmso به عنوان كنترل منفي استفاده گرديد. داده ها توسط آزمون t و واريانس يك طرفه anova آناليز شدند.يافته ها: ميانگين قطر هاله عدم رشد عصاره انهگزاني در غلظت 10 ميلي گرم بر ميلي ليتر بيشتر از قطر هاله عدم رشد آنتي بيوتيك نيستاتين براي كانديدا آلبيكنس و گلابراتا مي باشد (0.05>p). كمترين ميزان mic مربوط به عصاره انهگزاني براي كانديدا آلبيكنس و گلابراتا به ترتيب برابر با mug/ mul 15.62 و mug/ mul 31.25، كمترين ميزان mfc مربوط به عصاره انهگزاني براي كانديدا آلبيكنس و گلابراتا به ترتيب برابر با mug/ mul 31.25 و mug/ mul62.5 مي باشد.بحث و نتيجه گيري: عصاره هاي اين گياه اثر ضدقارچي مناسبي بر كانديدا آلبيكنس و گلابراتا نشان داد. بررسي احتمال كاربرد اين عصاره در درمان عفونت كانديدايي نياز به اتمام مطالعات تكميلي دارد.

Background : Because of the recurrence of Candida infections، their resistance to antifungal drugs during treatment، the known side effects of chemical drugs، and the existence of some studies that Onosma's genus contains derivatives of alkannin and shikonin that have antifungal properties، for the first time، the antifungal properties of Onosma Chlorotricum were compared with antibiotics. Materials and Methods: The plant was collected from Koohdasht in July of 2016، and after detection and drying، N- hexane، methanol، aqueous extracts of plants were prepared by soaking. The antifungal effects of different concentrations of extracts were studied by the disc diffusion method and determination of MIC (minimum inhibitory concentration of growth)، MFC (minimum fungicidal concentration) was done by microdilution method. Antibiotics fluconazole and nystatine were used as a positive control and DMSO was used as a negative control . Data were analyzed by t-test and one-way ANOVA. Results: The mean inhibitory diameter of growth of the hexane extract in concentration of 10 mg / ml was higher than nystatin antibiotic inhibitory diameter of growth for Candida albicans and Candida glaberata (P˂0.05). The lowest MIC for Candida albicans and Candida glaberata was respectively، 15.62 and 31.25 μg / μl، and the lowest MFC for Candida albicans and Candida glaberata was 31.25 and 62.5. μg / μl. Conclusion: Extracts of this plant showed antifungal effect on Candida albicans and glaberata. The possibility of using this extract in the treatment of Candida infection requires the completion of supplementary studies.
