چكيده فارسي :
زﻣﯿﻨﻪ وﻫﺪف: ﻫﺪف از اﯾﻦ ﭘﮋوﻫﺶ ﺑﺮرﺳﯽ اﺛﺮ 8 ﻫﻔﺘﻪ ﺗﻤﺮﯾﻦ اﺳﺘﻘﺎﻣﺘﯽ و رژﯾﻢ ﻏﺬاﺋﯽ ﺑﺮ ﺑﯿﺎن ژنPgc1α وUcp1 در ﺑﺎﻓﺖ ﭼﺮﺑﯽ ﻣﻮش ﻫﺎي ﻧﺮ ﭼﺎق و ﻧﺮﻣﺎل 6/C57BL ﻣﯽﺑﺎﺷﺪ. ﻣﻮاد و روﺷﻬﺎ: 48ﺳﺮﻣﻮش ﻧﺮ6/C57BLﺑﻪ ﺻﻮرت ﺗﺼﺎدﻓﯽ ﺑﻪ دو ﮔﺮوه ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ: 1. ﮔﺮوه ﺗﻐﺬﯾﻪ ﺷﺪه ﺑﺎ رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب، 2. ﮔﺮوه ﺗﻐﺬﯾﻪ ﺷﺪه ﺑﺎ رژﯾﻢ ﻏﺬاﯾﯽ ﮐﻢ ﭼﺮب )ﺑﻪ ﻣﺪت 12 ﻫﻔﺘﻪ(.ﺳﭙﺲ ﻫﺮ دو ﮔﺮوه ﭼﺎق و ﻧﺮﻣﺎﻟﺒﺮاﺳﺎس ﻣﯿﺎﻧﮕﯿﻦ وزﻧﯽ ﺑﺮاﺑﺮ ﺑﻪ ﭼﻬﺎر ﮔﺮوه ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ:رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب-ﺗﻤﺮﯾﻦ )HF-E(، رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب-ﺑﯽ ﺗﺤﺮك )-HF S(، رژﯾﻢ ﻏﺬاﯾﯽ ﮐﻢ ﭼﺮب-ﺗﻤﺮﯾﻦ )LF-E(، رژﯾﻢ ﻏﺬاﯾﯽ ﮐﻢ ﭼﺮب-ﺑﯽ ﺗﺤﺮك )LF-S(. ﻣﻮشﻫﺎي ﮔﺮوه ﺗﻤﺮﯾﻨﯽ ﺑﻪ ﻣﺪت 8 ﻫﻔﺘﻪ و 5 روز در ﻫﻔﺘﻪ، ﺑﺎ ﺳﺮﻋﺖ ﻣﺘﻮﺳﻂ 19 ﻣﺘﺮ در دﻗﯿﻘﻪ و ﺑﻪ ﻣﺪت 45 دﻗﯿﻘﻪ، ﺗﺤﺖ ﺗﻤﺮﯾﻦ روي ﺗﺮدﻣﯿﻞ ﻗﺮار ﮔﺮﻓﺘﻨﺪ.24 ﺳﺎﻋﺖ ﭘﺲ ازآﺧﺮﯾﻦ ﺟﻠﺴﻪ ﺗﻤﺮﯾﻨﯽ،ﻣﻮشﻫﺎ ﻗﺮﺑﺎﻧﯽ ﺷﺪﻧﺪ. ﺑﺎﻓﺖ ﭼﺮﺑﯽ ﺳﻔﯿﺪ زﯾﺮﭘﻮﺳﺘﯽ و ﭼﺮﺑﯽ ﻗﻬﻮه اي ﺟﺪا و ﺑﺮاي اﻧﺪازه ﮔﯿﺮي ﺑﯿﺎن Pgc1α وUcp1در ﻧﯿﺘﺮوژن ﻣﺎﯾﻊ ﻓﺮﯾﺰ ﺷﺪ و ﺑﺎ اﺳﺘﻔﺎده ازq-PCR ﺑﯿﺎن ژن ﮐﻤﯽ ﺳﺎزي وازآزﻣﻮن آﻧﺎﻟﯿﺰ وارﯾﺎﻧﺲ دوﻃﺮﻓﻪ در ﺳﻄﺢ 0/05˂pاﺳﺘﻔﺎده ﺷﺪ. ﻧﺘﺎﯾﺞ: ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد رژﯾﻢ ﭘﺮﭼﺮب ﻣﻨﺠﺮ ﺑﻪ اﻓﺰاﯾﺶ ﻣﻌﻨﯽ دار در ﺳﻄﻮح Pgc1α در ﺑﺎﻓﺖ ﭼﺮﺑﯽ ﺳﻔﯿﺪ زﯾﺮﭘﻮﺳﺘﯽ و ﭼﺎﻗﯽ ﻣﻨﺠﺮ ﺑﻪ ﮐﺎﻫﺶ ﻣﻌﻨﯽ دار آن ﺷﺪه اﺳﺖ. ﻋﻼوه ﺑﺮ اﯾﻦ، ﭼﺎﻗﯽ و ﺗﻤﺮﯾﻦ اﺳﺘﻘﺎﻣﺘﯽ ﻣﻨﺠﺮ ﺑﻪ ﮐﺎﻫﺶ ﻣﻌﻨﯽ دار ﺑﯿﺎن Pgc1α در ﺑﺎﻓﺖ ﭼﺮﺑﯽ ﻗﻬﻮه اي ﺷﺪه اﺳﺖ. ﻫﻤﭽﻨﯿﻦ ﺑﯿﺎن Ucp1 در ﮔﺮوهﻫﺎي ﭼﺎق ﺑﻪ ﻃﻮر ﻣﻌﻨﯽ داري ﮐﻤﺘﺮ از ﮔﺮوهﻫﺎي ﻧﺮﻣﺎل ﺑﻮده اﺳﺖ)0/05˂p(. ﻧﺘﯿﺠﻪ ﮔﯿﺮي: ﻧﺘﺎﯾﺞ ﻧﺸﺎن ﻣﯽدﻫﺪ ﮐﻪ ﺳﻄﻮح Pgc1α و Ucp1 در ﺑﺎﻓﺖ ﭼﺮﺑﯽ زﯾﺮﭘﻮﺳﺘﯽ و ﻗﻬﻮه اي ﻣﻮشﻫﺎي ﭼﺎق ﺑﻪ ﻃﻮر ﻣﻌﻨﯽ دار ﮐﺎﻫﺶ ﻣﯽﯾﺎﺑﺪ ﮐﻪ ﺑﯿﺎنﮔﺮ اﺧﺘﻼﻻت ﻣﺘﺎﺑﻮﻟﯿﮏ در اﺛﺮ ﭼﺎﻗﯽ ﻣﯽﺑﺎﺷﺪ. در ﺣﺎﻟﯽ ﮐﻪ ﺗﻤﺮﯾﻦ اﺳﺘﻘﺎﻣﺘﯽ ﻣﻨﺠﺮ ﺑﻪ اﻓﺰاﯾﺶ ﺑﯿﺎن Pgc1α و Ucp1 در ﺑﺎﻓﺖ ﭼﺮﺑﯽ ﺳﻔﯿﺪ زﯾﺮﭘﻮﺳﺘﯽ ﻣﯽﺷﻮد.
چكيده لاتين :
Background &Purpose: The aim of present study was to investigate the effect of eight-week endurance training and diet on the expression of Pgc1α and Ucp1 in adipose tissue of obese and normal C57BL / 6 mice.
Methodology: Accordingly, 48 male C57BL / 6 mice were randomly divided into two groups: high-fat diet group, low-fat diet group (12 weeks).Then, both obese and normal groups divided into four groups based on weight average: High-Fat diet Endurance (HF-E), High-Fat diet sedentary (HF-S), Low-Fat diet Endurance (LF- E), and Low-Fat diet sedentary (LF-S).Groups trained on treadmill for 8 weeks, 5 days a week, with 19 m/min for 45 minutes. 24h after the last training session, the mice were sacrificed. Subcutaneous and brown adipose tissue were isolated and frozen in liquid nitrogen to measure the expression of Pgc1α and Ucp1; using Q-PCR, for gene expression and two-way ANOVA (p˂0/05) as statistical analysis.
Results: The results showed that the high-fat diet caused significant increase in Pgc1α levels in the subcutaneous adipose tissue, while obesity resulted in a significant decrease in it. Additionally, obesity and training resulted in a significant reduction of Pgc1α expression in the brown adipose tissue. Also, the expression of Ucp1 in the obese groups was significantly lower than in the normal groups (p˂0/05). Also it was found that Pgc1α and Ucp1 levels were decreased significantly in the subcutaneous and brown adipose tissues of obese, indicating disorders due to obesity.
Conclusion: Training led to increase in the expression of Pgc1α and Ucp1 in the subcutaneous adipose tissue.
