Effects of Pseudomonas fluorescens on the disease severity, physiological, and biochemical traits in the tubers of potato cultivars infected with Rhizoctonia solani under greenhouse conditions

به منظور ارزيابي تأثير تيمار جدايه هاي باكتري Pseudomonas fluorescens بر ويژگي هاي فيزيولوژيكي، بيوشيميايي و شدت بيماري غده هاي بذري سيب زميني در حضور عامل بيماريزا Rhizoctonia solani، آزمايشي به صورت فاكتوريل در قالب طرح كاملاً تصادفي در سه تكرار انجام شد. تيمارهاي اين پژوهش شامل سه جدايه P. fluorescens UTPF5 (P5)، p. fluorescens UTPF68 (P68) و P. fluorescens UTPF74 (P74)، قارچ بيمارگر R. solani AG3 و ارقام سيب زميني شامل آگريا و سانته بود. صفات اندازه‌گيري شده در اين آزمايش شامل وزن خشك ريشه، طول استولون، وزن خشك استولون، وزن خشك غده، وزن تر غده، تعداد غده و تعداد استولون، شدت بيماري در غدههاي بذري و فعاليت آنزيم‌هاي GPX و β-1,3-glucanase و محتواي پرولين، مالون ديآلدهيد و قند محلول بود. نتايج نشان داد كه استفاده از تيمار باكتري توانست در حضور عامل بيماري زا تأثير معني‌داري بر صفات اندازه‌گيري شده داشته باشد و هم‌چنين شدت بيماري را نسبت به شاهد به طور معني‌داري كاهش دهد. تركيب تيماري P5 و رقم سانته نسبت به ساير تركيبات بر صفات اندازه گيري شده تأثير بيشتري داشت، به‌طوري كه بيشترين وزن خشك ريشه، طول استولون، وزن خشك استولون، وزن خشك غده، وزن تر غده و تعداد استولون در اين تيمار به دست آمد و نسبت به شاهد به‌ترتيب افزايش 0/62، 0/86، 1/40، 2/31، 1/37 و 0/63 برابري مشاهده شد. استفاده از تيمار P5باعث افزايش فعاليت سيستم آنتياكسيداني آنزيم گلوتاتيون پراكسيداز (GPX) و بتا 1 و 3 گلوكاناز (β-1,3-glucanase)، محتواي پرولين و قند محلول به ترتيب به ميزان 1/32، 0/61، 2 و 2/6 برابر و از طرف ديگر كاهش پراكسيداسيون ليپيد به ميزان 57 درصد نسبت به شاهد در شرايط حضور عامل بيماري زا شد.
چكيده لاتين :
To evaluate the effects of Pseudomonas fluorescens on potato production in the presence of Rhizoctoniasolani,a three replicated greenhouse experiment was conducted in a completely randomized factorial design containing three strains of Pseudomonas fluorescens includingUTPF5(P5),P. fluorescensUTPF68 (P68) and P. fluorescens UTPF74(P74), Rhizoctonia solani AG3 and Agria and Sante cultivars of potato provided by the seed and plant certification and registration institute. Disease severity, root dry weight, stolon length and dry weight, tubers number, dry weight and fresh weight, Antioxidant activity (GPX, β-1, 3-glucanase), proline content, MDA and soluble sugar content were determined in different treatments. The results showed that Pseudomonas fluorescens treatment had a significant effect on all traits in the presence of fungus(pathogen-infected condition). The fungal virulence was also significantly decreased in this treatment compared to the control. P5 and Santevariety combined treatment showed highly significant effect on the measured traits than the other combined treatments, so that the highest amounts of dry weight of root, stolon length and dry weight, tubers dry and fresh weight and the number of stolon were observed in this treatment with 0.62, 0.86, 1.40, 2.31, 1.37 and 0.63 folds increase respectively compared to the control. Results also indicated that in P5 treatment Antioxidant enzyme activity, (GPX, β-1, 3-glucanase), proline and soluble sugar content were increased by 1.32, 0.61, 2 and 2.6 and folds respectively and lipid peroxidation was decreased by 53% in comparison with the control.
