چكيده فارسي :
ﻫﺪف از اﻧﺠﺎم ﺗﺤﻘﯿﻖ ﺣﺎﺿﺮ ﺑﺮرﺳﯽ اﺛﺮ ﯾﮏ دوره ﺗﻤﺮﯾﻨﺎت ﻫﻮازي ﺑﺮ ﺑﯿﺎن PI3K / AKT1و ﺣﺴﺎﺳﯿﺖ اﻧﺴﻮﻟﯿﻦ در ﻫﭙﺎﺗﻮﺳﯿﺖ ﻫﺎي ﮐﺒﺪي رت ﻫﺎي وﯾﺴﺘﺎر ﭼﺎق ﻣﯽ ﺑﺎﺷﺪ. ﺟﺎﻣﻌﻪ آﻣﺎري در اﯾﻦ ﺗﺤﻘﯿﻖ را ﮐﻠﯿﻪ رت ﻫﺎي ﻧﺮ وﯾﺴﺘﺎر ﺣﯿﻮان ﺧﺎﻧﻪ داﻧﺸﮕﺎه ﻋﻠﻮم ﭘﺰﺷﮑﯽ ﺑﻘﯿﻪ اﷲ اﯾﺮان ﺗﺸﮑﯿﻞ داده ﺑﻮدﻧﺪ.آنﻫﺎ ﻃﯽ ﻫﺸﺖ ﻫﻔﺘﻪ رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮ ﭼﺮب ﭼﺎق ﺷﺪﻧﺪ. 16 ﺳﺮ رت وﯾﺴﺘﺎر ﮐﻪ ﻫﻤﮕﯽ از وﯾﮋﮔﯽ ﻫﺎي ﻓﯿﺰﯾﮑﯽ و ﺳﻨﯽ ﻣﺸﺎﺑﻬﯽ ﺑﺮﺧﻮردارﺑﻮدﻧﺪ. ﺑﻪ ﺷﯿﻮه ﺗﺼﺎدﻓﯽ در 2 ﮔﺮوه ﭼﺎق )ﮐﻨﺘﺮل و ﺗﻤﺮﯾﻦ ﻫﻮازي( ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﺑﺮاي اﯾﺠﺎد ﭼﺎﻗﯽ، از رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب ﺑﻪ ﻣﺪت 8 ﻫﻔﺘﻪ اﺳﺘﻔﺎده ﺷﺪ. ﺟﻬﺖ ﺗﻬﯿﻪ ﻏﺬاي ﭘﺮﭼﺮب ﺑﻪ ﻏﺬاي اﺳﺘﺎﻧﺪارد ﻣﻮش ﻫﺎي ﺻﺤﺮاﯾﯽ ﮐﻪ از ﺷﺮﮐﺖ ﺧﻮراك ﭘﺎرس دام ﺧﺮﯾﺪاري ﮔﺮدﯾﺪ 1% ﭘﻮدر ﮐﻠﺴﺘﺮول و 1% روﻏﻦ ذرت 100% ﺧﺎﻟﺺ، اﺿﺎﻓﻪ ﺷﺪ. رت ﻫﺎ در آزﻣﺎﯾﺸﮕﺎه ﺣﯿﻮاﻧﺎت در ﺷﺮاﯾﻂ ﮐﻨﺘﺮل ﺷﺪه ﻧﻮر )ﺷﺎﻣﻞ 12 ﺳﺎﻋﺖ روﺷﻨﺎﯾﯽ و 12 ﺳﺎﻋﺖ ﺗﺎرﯾﮑﯽ، ﺷﺮوع روﺷﻨﺎﯾﯽ 6 ﻋﺼﺮ و ﺷﺮوع ﺧﺎﻣﻮﺷﯽ 6 ﺻﺒﺢ و دﻣﺎ 22±3 ﺳﺎﻧﺘﯽ ﮔﺮاد و رﻃﻮﺑﺖ ﺣﺪود 30 ﺗﺎ 60 درﺻﺪ ﺑﻮد( ﻧﮕﻬﺪاري ﺷﺪﻧﺪ. اﺳﺘﻔﺎده از رژﯾﻢ ﻏﺬاﯾﯽ ﭘﺮﭼﺮب ﺗﺎ اﻧﺘﻬﺎي ﻣﻄﺎﻟﻌﻪ اداﻣﻪ داﺷﺖ. ﺑﺮﻧﺎﻣﻪ ﺗﻤﺮﯾﻨﯽ در ﮔﺮوه ﻫﻮازي ﺑﺮاي ﻣﺪت 6 ﻫﻔﺘﻪ ﺑﻪ ﺗﻌﺪاد 5 ﺟﻠﺴﻪ در ﻫﻔﺘﻪ ﺑﺎ اﻓﺰاﯾﺶ ﺗﺪرﯾﺠﯽ ﺳﺮﻋﺖ )18 اﻟﯽ 22 ﻣﺘﺮ ﺑﺮ دﻗﯿﻘﻪ( و زﻣﺎن )10 اﻟﯽ 55 دﻗﯿﻘﻪ در ﻗﺎﻟﺐ دوﯾﺪن روي ﺗﺮﯾﺪﻣﯿﻞ( اﻧﺠﺎم ﮔﺮﻓﺖ. ﻫﻤﻪ رت ﻫﺎ 48 ﺳﺎﻋﺖ ﭘﺲ از آﺧﺮﯾﻦ ﺟﻠﺴﻪ ﺗﻤﺮﯾﻨﯽ ﺗﺸﺮﯾﺢ ﺷﺪﻧﺪ. ﻧﺘﺎﯾﺞ ﺗﺤﻘﯿﻖ ﻧﺸﺎن داد ﮐﻪ 6 ﻫﻔﺘﻪ ﺗﻤﺮﯾﻦ ﻫﻮازي ﺑﻪ ﮐﺎﻫﺶ ﻣﻌﻨﯽ دار ﮔﻠﻮﮐﺰ ﻧﺎﺷﺘﺎ در رت ﻫﺎي وﯾﺴﺘﺎر ﭼﺎق ﻣﻨﺠﺮ ﺷﺪ. و از ﻃﺮﻓﯽ ﻣﻨﺠﺮ ﺑﻪ اﻓﺰاﯾﺶ ﺑﯿﺎنPI3K / AKT1 و ﺣﺴﺎﺳﯿﺖ اﻧﺴﻮﻟﯿﻦ در ﺑﺎﻓﺖ ﮐﺒﺪي رت ﻫﺎي وﯾﺴﺘﺎر ﭼﺎق ﺷﺪ ) 0/05 P≤(.
چكيده لاتين :
The purpose of the present study was to investigate the effect of aerobic exercises on AKT1/ PI3K expression and Insulin sensitivity in liver Hepatocyte of obese Vistar rats. The statistical population consisted of all male Wistar rats of Baqiyatallah University of Medical Sciences in Iran. To create obesity, a high-fat diet was used for 8 weeks. 16 obese rat having similar condition were selected and randomly divided into 2 obese groups (control and aerobic exercise). To prepare a high-fat diet, 1% cholesterol powder and 1% pure corn oil were added to the standard diet of rats purchased from Parsdam Food Company. Rats in animal laboratory under controlled light conditions (12 hours light and 12 hours dark, start of lighting 6 pm and start of extinction 6 am and temperature 22 ± 3 ° C and humidity about 30 to 60%) were maintained. The use of a high-fat diet continued until the end of the study. The aerobic exercise program was performed for 6 weeks in 5 sessions per week with a gradual increase in speed (18 to 22 meters per minute) and time (10 to 55 minutes in the form of running on a treadmill). All rats 48 hours after the last training session were autopsied. The results showed that 6 weeks of aerobic training led to a significant reduction in fasting glucose in obese Wistar rats. On the other hand, it increased AKT1 / PI3K expression and insulin sensitivity in liver tissue of obese Wistar rats (P≤0.05).
