اثرات مصرف جداگانه پروبيوتيك و تركيب با پربيوتيك جيره بر ساختار بافت‌شناسي روده و پارامترهاي بيوشيميايي خون بچه ماهي صبيتي (Sparidentex hasta)

Effects of using single probiotic and combination with dietary prebiotic on intestine histological structure and blood biochemical parameters of Sobaity (Sparidentex hasta) fingerling

پرورش متراكم آبزيان و توليد تجاري نياز به بهبود شرايط بهداشتي ماهي دارد و پروبيوتيك‌ها و پرييوتيك‌ها به‌عنوان گزينه مناسب جهت رسيدن به اين هدف در نظر گرفته مي‌شوند. هدف از انجام اين مطالعه بررسي اثرات جداگانه و تركيبي پروبيوتيك با پربيوتيك جيره بر بافت‌شناسي روده و پارامترهاي بيوشيميايي خون بچه ماهي صبيتي (Sparidentex hasta) است. براي اين منظور تعداد 180 قطعه ماهي با ميانگين وزني 0/8±35/14 گرم از پژوهشكده آبزي‌پروري جنوب كشور تهيه شد. اين مطالعه در قالب يك طرح كاملاً تصادفي با 4 تيمار و 3 تكرار (15 قطعه ماهي به ازاي هر تكرار) در داخل مخازن فايبرگلاس 300 ليتري انجام شد. بچه ماهيان به مدت 8 هفته با جيره هاي فاقد پروبيوتيك و پربيوتيك (گروه شاهد)، حاوي 106×1 CFU پروبيوتيك (Lactobacillus plantarum) در هر گرم غذا (تيمار 1)، 0/5 و 1 درصد زايلواوليگوساكاريد + 106×1 CFUپروبيوتيك در هر گرم غذا ( تيمار 2 و 3) در حد سيري ظاهري تغذيه شدند. در پايان آزمايش نمونه‌هاي بافت روده و خون جمع آوري شدند. نتايج نشان داد كه تركيب پروبيوتيك و پربيوتيك جيره عملكرد ساختار بافتي روده ماهيان را تحت تاثير قرار نمي‌دهد (0/05>P). در پايان آزمايش فعاليت پارامترهاي بيوشيميايي خون (گلوكز، تري‌گليسريد، كلسترول، HDL، LDL، VLDL) اختلاف معني‌داري بين گروه شاهد و ساير تيمارهاي آزمايش نشان نداد (0/05>P). به‌طور كلي، اين مطالعه نشان داد كه افزودن تركيب پروبيوتيك با پربيوتيك در مقادير استفاده شده اثرات معني‌داري بر بافت‌شناسي روده و پارامترهاي بيوشيميايي خون ماهي صبيتي ندارد. با اين حال افزودن جداگانه پروبيوتيك افزايش طول ويلي را به دنبال داشت.

Intensive aquaculture and commercialization production are believed to be needed for improving fish health status and probiotics and prebiotics are candidates to achieve this goal. The aim of this study was to evaluate the single and combined effects of dietary probiotic with prebiotic on intestine histology and blood biochemical parameters of Sobaity fingerling (Sparidentex hasta). For this purpose, a total of 180 Sobaity fingerlings were prepared with an average weight of 35.14±0.8g from the Mariculture Research Station of South Iranian Aquaculture Research Center. This study was carried out in a completely randomized design with four treatments and three replications (15 fish per each replication) in fiberglass tanks with 300 liters volume. Fish were fed with feed containing without supplementation (control group), probiotic 106 CFU/g feed (treatment 1), 0.5 and 1 percent prebiotic 106 CFU/g feed (treatment 2 and 3) at satiation for a period of 8 weeks. At the end of the experiment, the intestine and blood samples were collected. The results showed that combination of dietary probiotic with prebiotic did not change intestine histology of Sobaity fish (P>0.05). At the end of the experiment, the dietary prebiotic and probiotic also did not change blood biochemical parameters (glucose, triglycerides, cholesterol, HDL, LDL and VLDL) (P>0.05). This study showed that combination of probiotic with prebiotic at the used amounts had no significant effects on intestine histology and blood biochemical parameters of Sobaity fish. However, single supplementation of probiotic causes the elevation of villi length.
