چكيده فارسي :
ﺑﯿﻦ ﺑﯿﻤﺎري ﺻﺮع و ﮐﺎﻫﺶ ﺣﺎﻓﻈﻪ روزاﻧﻪ، ﯾﺎدﮔﯿﺮي و اﺧﺘﻼل رﻓﺘﺎري ارﺗﺒﺎط وﺟﻮد دارد ﻟﺬا در اﯾﻦ ﺗﺤﻘﯿﻖ ﺗﺎﺛﯿﺮ ﻣﻨﺘﻮل ﺑﺮ ﯾﺎدﮔﯿﺮي و ﺣﺎﻓﻈﻪ ﻓﻀﺎﯾﯽ و ﺑﯿﺎن ژن ﮔﯿﺮﻧﺪه ﮔﺎﺑﺎ در ﻣﻮش ﻫﺎي ﺻﺮﻋﯽ ﻣﻮرد ﺑﺮرﺳﯽ ﻗﺮار ﮔﺮﻓﺖ. در اﯾﻦ ﺗﺤﻘﯿﻖ 60 ﺳﺮ ﻣﻮش ﺻﺤﺮاﯾﯽ ﺑﻪ 6 ﮔﺮوه 10 ﺗﺎﯾﯽ ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ، ﺷﺎﻣﻞ ﮔﺮوه ﻫﺎي ﮐﻨﺘﺮل، اﺗﺎﻧﻮل، PTZ37/5 mg/kg) (، ﻣﻨﺘﻮل )mg/kg 30( و ethanol+pTZوMenthol+PTZ. ﮐﯿﻨﺪﻟﯿﻨﮓ ﺷﯿﻤﯿﺎﯾﯽ ﻃﯽ ﯾﮏ دوره 28 روزه ﺑﺎ ﺗﺰرﯾﻖ 14 ﻧﻮﺑﺖ ﭘﻨﺘﯿﻠﻦ ﺗﺘﺮازول ﺑﺼﻮرت ﯾﮏ روز در ﻣﯿﺎن اﯾﺠﺎد ﺷﺪ، ﺳﭙﺲ ﯾﺎدﮔﯿﺮي و ﺣﺎﻓﻈﻪ ﻓﻀﺎﯾﯽ ﺑﻪ وﺳﯿﻠﻪ دﺳﺘﮕﺎه ﻣﺎز آﺑﯽ ﻣﻮرﯾﺲ ﻣﻮرد آزﻣﺎﯾﺶ ﻗﺮار ﮔﺮﻓﺖ. ﺳﭙﺲ ﺣﯿﻮاﻧﺎت ﺑﯿﻬﻮش ﺷﺪه و ﻣﻐﺰ آﻧﻬﺎ ﺑﺮاي ﺑﺮرﺳﯽ ﺑﯿﺎن ژن GABBR2 ﺟﺪا ﮔﺮدﯾﺪ، ﺑﺮاي آﻧﺎﻟﯿﺰ آﻣﺎري داده ﻫﺎ، از آزﻣﻮن ANOVA ﯾﮑﻄﺮﻓﻪ و دوﻃﺮﻓﻪ و آزﻣﻮن ﺗﻮﮐﯽ در ﺳﻄﺢ ﻣﻌﻨﯽ داري P<0/05 و ﺑﺮاي ﺑﯿﺎن ژن ﻧﯿﺰ از ﺗﺴﺖ دﻟﺘﺎ ﺳﯽ ﺗﯽ اﺳﺘﻔﺎده ﺷﺪ. PTZ ﯾﺎدﮔﯿﺮي و ﺣﺎﻓﻈﻪ را ﮐﺎﻫﺶ داد و ﻣﻨﺘﻮل ﺗﻮاﻧﺴﺖ از اﯾﻦ ﻧﻘﺺ ﺟﻠﻮﮔﯿﺮي ﮐﻨﺪ و ﯾﺎدﮔﯿﺮي و ﺣﺎﻓﻈﻪ را اﻓﺰاﯾﺶ دﻫﺪ، ﻫﻤﭽﻨﯿﻦ ﻣﻨﺘﻮل ﺗﻮاﻧﺴﺖ ﺑﯿﺎن ژن GABBR2 را در ﻣﻮش ﻫﺎي ﺻﺮﻋﯽ اﻓﺰاﯾﺶ دﻫﺪ. ﭘﯿﺶ درﻣﺎﻧﯽ ﺑﺎ ﻣﻨﺘﻮل در ﻣﻮش ﻫﺎي ﺻﺮﻋﯽ ﺷﺪه ﺑﺎ ﭘﻨﺘﯿﻠﻦ ﺗﺘﺮازول، ﺑﻄﻮر ﻣﺤﺴﻮﺳﯽ از ﺑﺮوز ﺻﺮع ﺟﻠﻮﮔﯿﺮي ﮐﺮد. ﻣﻨﺘﻮل ﯾﺎدﮔﯿﺮي، ﺣﺎﻓﻈﻪ ﻓﻀﺎﯾﯽ و ﺑﯿﺎن ژن ﮔﯿﺮﻧﺪه ﮔﺎﺑﺎ را در ﻣﻮش ﻫﺎي ﺻﺮﻋﯽ ﺷﺪه اﻓﺰاﯾﺶ داد، ﺑﻨﺎﺑﺮاﯾﻦ اﺣﺘﻤﺎﻻ ﻣﻨﺘﻮل ﻣﯽ ﺗﻮاﻧﺪ ﯾﮏ اﻧﺘﺨﺎب ﻣﻨﺎﺳﺐ ﺟﻬﺖ درﻣﺎن و ﮐﺎﻫﺶ اﺧﺘﻼﻻت ﯾﺎدﮔﯿﺮي و ﺣﺎﻓﻈﻪ ﻧﺎﺷﯽ از ﺻﺮع ﺑﺎﺷﺪ.
چكيده لاتين :
There is a relationship between epilepsy and decreased daily memory, learning and behavioral disorders, so in this study, the effect of menthol on learning and spatial memory and GABA receptor gene expression in epileptic rats was investigated. Materials and Methods: In this study, 60 male rats were randomly divided into 6 groups of 10, including control , ethanol, pTZ (37.5 mg / kg), menthol (30 mg / kg) and ethanol + PTZ and Menthol + PTZ groups Chemical kindling was performed over a 28-day period with 14 injections of pentylenetetrazole every other day, then spatial learning and memory were tested by the Morris water maze .Then, the animals were anesthetized and their brains were isolated for GABBR2 gene expression. For statistical analysis of data, one-way and two-way ANOVA and Tukey test were used at a significant level of P <0.05. Delta CT test was used to express the gene. Finding: PTZ decreased learning and memory and menthol was able to prevent this defect and increase learning and memory, also menthol was able to increase GABBR2 gene expression in epileptic rats. Conclusion: Pretreatment with menthol significantly reduced the incidence of epilepsy in epileptic rats with pentylenetetrazole. Menthol increased learning, spatial memory, and GABA receptor gene expression in epileptic rats, Therefore, menthol may be a suitable choice for the treatment and reduction of learning and memory disorders caused by epilepsy.
