THE EFFECT OF EPIDERMAL GROWTH FACTOR ON DEVELOPMENT OF MOUSE MORULAE IN VITRO

The murine pre-implantation embryos express variety of growth receptors and their ligands. One of the factors is epidermal growth factor (EGF) which expresses through pre-implantation embryo and it is also present in oviductaI fluid. EGF stimulates cellular proliferation and differentiation and acts as a mitogen that promotes mitosis and differentiation of murine trophoectoderm. The purpose of this research was to study the effects of EGF on mouse morulae development in vitro and determination of the suitable dose. Mouse morula embryos were obtained from pregnant female NM RI mice after superovulation. The embryos were divided into four groups; control and experimental 1,2 and 3. The control group embryos were cultured on T6 medium, the first experimental an T6 + EGF (10 ng/ml), the second cxl- on ʹlʹ6+EGF (4 ng/ml) and the last experimental groups embryos were cultured on T6+EGF (1 ng/ml). Developmental rates were recorded daily until 96h. Data was analyzed using x2 test. The results showed that $3.2% of exp. 1 group reached hatching blastocyst stage while 70.6% of control and 67% and 66.7% of exp. 2 and 3 groups reached this developmental stage and the differences were statistically significant. There was no significant difference between degeneration rate of exp l group. and control group but the other two exp. groups showed high rate of degeneration. The results indicated that adding of EGF (10 ng/ml) to culture medium can enhance the development of mouse morulae until hatching blastocyst stage .
