Bacteriological Study of Post Space Preparation Timing on Apical Seal

Introduction: Corona) seal during treatment is necessary, because could prevent penetration bacteria, saliva and bacterial products to root canal system and attach to periodontal tissue. Purpose: The purpose of this study was bacteriological evaluation of the post space preparation on apical seal. Materials and Methods: A total 36 intact human premolar with straight roots were selected for this study and 32 teeth were prepared with step back technique and filled by lateral condensation of gutta percha plus AH26. All teeth divided in to two groups. Group one included 14 teeth that post space was immediately prepared and group two included 14 teeth that post space was prepared after one week. Both groups including 2 teeth after step back without gutta percha plus AH26, as positive controls and 2 intact teeth as negative controls. Proteus vulgaris was developed based on Gish model system for bacteriological of coronal leakage in 40 days. Results: All specimens of experimented group one did not show penetration of Proteus vulgaris, but 2 specimens of group 2 showed penetration of P.vulgaris. The exact fisher test revealed no significant statistical difference between two groups (p>0.05). Conclusion: This study suggested no difference between immediate and delayed post space preparation on apical seal in 40 days.
