Anti-Dog Immunoglublolins is equivalent to human Coombs reagent that could be useful for diagnostic-academic purposes including Coombs Wright test in dogs affected by brucellosis due to Brucella canis.However, anti-dog immunoglubolins have application in diagnosis and instruction of Direct Coombs, Indirect Coombs and Double Diffusion tests to the students of Medical, Pharmacy and Veterinary Sciences.Anti-Dog Igs conjugated with peroxidase also has application in designing of ELISA kits in order to diagnosis of cutaneous and Visceral Leishmaniasis.The goal of This research is taking more step toward Iran self sufficiency by spending the least economical expenses. For production of polyclonal antibody against dog Igs providing of an enriched enriched fraction of dog Igs for immunization is required. Since dog Igs are naturally soluble, to stabilize them in the body, they should be mixed with adjuvant and injected subcutaneously and intramuscularly to rabbit. After immunization of the animal at a certain time period for producing enriched serum of instant antiimmunoglobulins, rabbit was bleeded.For evaluation of anti-dog Igs, Ouchterlony and Single Radial Immunodiffusion or SRID tests were used. In these tests interaction between antigen and antibody were shown by sharp line and clear ringed precipitation respectively .For exact measurment of anti-dog Igs titr, a Designed ELISA test was used.The final titer of immunized rabbit serum was determined 4000.The result of Ouchterlony test indicated that a clear precipitation line appears between antigen and antibody wells due to well immunization of the rabbit.The results of SRID test confirmed the Ouchterlony test result due to formation of clear precipitation ring between diffused dog Igs in the wells with anti-dog Igs in the gel.Finally, the results of titration with the designed ELISA confirmed the results of Ochterlony and SRID tests due to the appearance of 4000 titer that shows the economical benefit of the product.
